En réponse à une protéine étrangère, le système réticulo-endothélial peut produire plusieurs anticorps qui peuvent posséder une affinité pour des fractions différentes de cette protéine antigénique. On peut mettre en évidence ces composants différents en laissant l'antigène et le sérum qui contient les anticorps diffuser l'un vers l'autre à l'intérieur d'un gel. L'endroit où ils se rencontrent se marque d'une ligne blanche de précipité après 24 à 48 heures d'incubation à 37°.
